Fire Rescue Victoria (FRV) responded to a fire on Donnybrook Road, Norlane, around 1:41 a.m. after reports of a vehicle and shop ablaze.
Firefighters arrived within six minutes, confirming no occupants and containing the fire to the shop with CFA support.
The blaze was under control by 2:12 a.m., with police and SES also on scene.
Detectives from Taskforce Lunar believe the fire was caused by offenders ramming a vehicle into the tobacconist shop and setting it alight.
A crime scene has been established, and investigators are examining links to other recent incidents.
